人肝癌细胞与人胚肝细胞核基质的免疫相关性 被引量:2

摘要 核基质是活性基因锚定的结构部位。本文用高盐缓冲液提取制备人肝癌细胞及人胚肝细胞核基质;以此作为抗原,制备出相应抗血清;用以探讨人肝癌细胞核基质的免疫特异性及其与人胚肝细胞在免疫酶标定位上的相关性。结果表明:人肝癌细胞核基质及人胚肝细胞核基质产生的抗血清特异地与各自对应抗原呈免疫酶标阳性反应,并主要局限在核与核膜上。而与其它对照组织如:正常人肝、大鼠肝、人乳腺癌及人结肠癌的核基质呈阴性反应。人胚肝核基质抗血清与人肝癌核基质免疫酶标反应中呈明显的交叉反应。提示人胚肝与人肝癌在细胞分化及基因表达的调控蛋白等方面存在一定的免疫相关性。 The nuclear matrix was extractedfrom nuclei in human f6t3l Iiverand hepatic carcinoma by high salt solution。BALB/c mice were im munized by thenuclear matrix of human fetal liver or hepatoma。The sera from immunized animals wereused as probe to detect the relation between humarn fetal liver and hepat carcinoma bymeans of immuno-histochemistry。The results show that aritisera of nuclear matrix ofhuman leta1 liver or hepatic carcinoma reacted speclfically to their targed tisque,but notto tiSsues from normal human liver, rat liver, human colon cancer and breast tumor。Moreover,there are cross-reaction between the antisera of human fetal liver and hepaticcarcinoma,noma,suggesting there must be certain relation between both of them inhistogenesis and regulation of gene expression。
