平阳霉素（A5）与抗人食管癌非组蛋白抗体交联物对人食管癌Eca－109细胞株的体外抑制效应

CYTOSTATIC EFFECTS OF NHPIgG-A5 CONJUGATE ON THE HUMAN ESOPHAGEAL CANCER CELL LINE Eca-109 IN VITRO

我们以非组生白质（ＮＨＰ）抗体作为抗癌药物平阳霉素（Ａ５）的载体，制备ＮＨＰＩｇＧＡ５交联物。应用（１）细胞培养隔日计数示生长曲线；（２）以Ｆｅｕｌｇｅｎ法示细胞ＤＮＡ相对含量；（３）３Ｈ－ＴｄＲ掺入实验、比较了ＮＨＰＩＧ－Ａ５、ＮＨＰＩｇＧ及Ａ５对人食管癌细胞株ＥＣａ１０９的体外抑制效应，四者比较，ＮＨＰＩｇＧ－Ａ５人食管癌Ｅｃａ－ｌ０９细胞株有较强的抑制效应。

Bleomycin A5 was conjugated to anti-NHPlgG by sodium periodateoxidation method.The cytostatic effect of NHP、 NHPIgG-A5 conjugate. free A5 andCEAIgG-A5 conjugate on the human esophageal cancer cell line: ECAa-109 cell, wasstueied : ① the growth curve of cells, ② the relative content of DNA of cells; ③ inc-orporation of 8H-TDR into cells. The results Showed that the four agents expressedcytostatic activity against the Eca-109 cell line. The most effective agent was NHP-IgG-A5. It is concluded that the cytostatic effect of NHPIgG-A5 conjugate was super-ior to that of CEAIgG-A5 conjugate, free A5 and NHPIgG alone.
