摘要
以生物素标记的EB病毒BamHI酶切w片段为探针,原位检测人鼻咽癌等石蜡切片中的EB病毒DNA,结果发现:在部分标本的癌细胞胞浆中和非癌的鼻咽增生上皮细胞中,鼻咽粘膜下小腺体及唾液腺细胞浆中,存在内源性的亲和素结合活性,因此产生假阳性信号。文章介绍了设立相邻切片的无探针对照来区分真伪的方法,并对生物素标记探针原位杂交技术的适用范围进行了讨论。
In the experiment of in situ detection of EBV-DNA on routine paraffin sections of nasopharyngeal carcinoma biopsies’it was discovered that some cells in some biopsies showed endogenous avidin binding activity which resulted in pseudo-signals.The method used to distinguish the false and true positive signals was introduced and the applicable scope of biotinylated probe on in situ hybridization was discussed.
