摘要
利用PCR技术从人外周血T细胞cDNA文库中扩增出人IL-9的cDNA,将其连入大肠杆菌表达载体后,可在大肠杆菌高效表达出人重组IL-9融合蛋白,表达量占菌体蛋白的25%左右,有明显的刺激正常人骨髓细胞集落形成的活性。
The
cDNA coding region for human interleukin-9(IL-9)Was amplified from a
human T cell cDNAlibrary with PCR technique. The PCR products were
subcloned into a E. coli expression、vecor and human recombinant
IL-9 fusion protein was expressed with high ejjiciency in E. coli .Up
to 25%of the total bacterial proteins was the recombinant IL-9.
Biological activity analysis showed that the exp- ressed
IL-9stimulated increased colony-formation of normal human bone marrow
cell.
