摘要
采用电镜原位杂交法,观察6例急性白血病免疫球蛋白重链(IgH)基因重排,结果发现,5例急性淋巴细胞白血病(ALL)阳性,表明均为B-ALL;1例急性粒单细胞白血病(AML-M4)阴性。在阳性细胞核内,代表IgH基因重排的胶体金颗粒成群分布,该技术把核酸探针杂交与超微形态学相结合,在基因水平确定白血病细胞类型,尤其对缺乏特异免疫表型的白血病更有应用价值。
Immunoglobul n heavy chain(IgH)gene rearrangements were observed in
six patients with acute leukemia using electron microscopic in situ
hybridization,The results indicated that five cases with acute
lymphocytic leukemia (ALL) were positive,and one case with AML- N4
was negative. Five caseswith ALL were B- ALL. In positive
nuclei,colloid gold particles which represented IgH gene rearran-
gem ent distrlbuted in clusters. This technique combined nucleic acid
probe hybridization with ultrastru-ctural morphology,and it was
possible to identify leukemic cells at the gene level,probably more
imp-ortant in acute leukemia cases that lack special
immunophenotype.
