aving an Asp→Ala substitution in aa144 of the major S, a new HBV mutant wasidentified from the blood sample of a persistently infected patient #105, who was born from a carrier mother positive both in HBsAg and HBeAg. Patient #105 had been immunized with HBV vaccine after parturition under a conventional 0、1、6 SCHEDULE, BUT FAILED IN BEING PRO-tected. This major S aa144Asp→Ala HBV mutant has been detected both in the blood samples of patient # 105 at 1 and 4 years of age which indicated that this new HBV mutant is compe- tent in replication. As the aa144 is just next to the aa145,which was reported to be criticalfor the“a”epitope, the substitution of Asp to Ala in aa144 should be important also also for“a”and this HBV major S mutant might be a new escape mutant which led to the failure of pro-tection by HBV vaccine composed of wild type S antigen.
