epatitis B Virus(HBV)DNA replication takes place inside the nucleocapsid via reversetranscription.It was considered that modified abnormal core proteins might compete for normal core proteins in the formation of functional nucleocapsids and hence,block thereplication of HBV DNTo test the above hypothesis, we constructed five recombinant plasmids which could separately express normal and modified HBV core proteins withinHepG2 cells.The constructs were assayed for their potential to inhibit HBV replication bycotransfection with an equal amount of HBV DNA dimer plasmid (p177,which has been proved to be able to replicate in HepG2 cells after transfection )into HepG2 cells.The replication and ex-pression of HBV DNA were determined 5 days post-transfection by Southern blot analysis of DNAs extracted from extracellular virions and detection of the HBsAg and HBeAg levels in cul-ture medium using Abbott-ELISA Kit.Results showed that modified core proteins could in-hibit HBV gene replication and expression.This study indicates that modified core proteins might develop into a novel group of anti-HBV agents.
