山莨菪碱对心肌肌膜Na^+-Ca^(2+)交换蛋白活性呈剂量依赖性的抑制作用,而异搏定不抑制Na^+-Ca^(2+)交换,高浓度反而呈轻度促进作用,内毒素则与山莨菪碱的作用相反。提示抑制Na^+-Ca^(2+)交换可能是山莨菪碱发挥其细胞保护作用的机制之一,
In order to study the active mechanism of anisodamine at cellular level, we compared the effect of anisodamine, verapamil and endotoxin on Na+-Ca2+ exchange activity of canine cardiac sarcolemma in reconsituted liposo-mes. The results showed that anisodamine dose-dependently inhibited the activity of Na + -Ca2+ exchange, however, verapamil did not inhibit it at low dose, or slightly stimulated it at high dose. contrary to anisodamine, endotoxin was a dose-dependent stimulator of Na+- Ca2+ exchange. The present study suggests that inhibition to Na- + Ca2+ exchange may be one of the cyto-protection mechanisms of anisodamine in disease therapy.
