摘要
用0.9%的碘乙酰胺溶液灭活啤酒酵母S.cercvisiaeMA的原生质体与赖氨酸缺陷型啤酒酵母S.cerceisiaeCAlys-原生质体以PEG为融合剂诱导融合。在基本培养基平板上选择到数株融合子再生菌株。融合株经过传代培养,细胞形态、DNA含量及细胞内可溶性蛋白质的聚丙烯酰胺凝胶电泳分析,确证是两亲本的融合株。
The paper presents the
results of protoplast fusion betweeniodoacetamide-inactivated protoplast of S.cerevisiae MA
and protoplast of lysine-requiring S.cerevisiae CA induced by polyetheneglycol(PEG,MW 6000).
The concentration of iodoacetamide solution was 0.9%.The protoplast was inactivated for 10
minutes at 30℃.Fusants were picked out on selective medium HYMM,with a fusion rate of 1.82
×10-5.The fusants were cultivated continuously through 10 generations on mdeium YMM.After
obtaining the stable fusants from YMM,comparative studies on cell size,cell morphology,DNA
content per cell,fermentation indexes and eletrophoretic soluble protein patterns got by PAGE
between the fusants and their parental strains were made.These data confirm that the fusants
are heterozygotes of both parental strains.
