胆红质的反应浓度因素在双波长全自动生化分析仪中的作用

摘要 胆红质的测定在临床具有重要的意义,但由于胆红质不稳定,胆红质的标准液是造成测定误差的一个重要原因。且标准液昂贵,又不易保存,为了解决这一问题,把胆红质标准用双波长全自动生化分析仪连续测定,通过反应吸光度和标准液含量求出反应浓度因素。在同一批试剂中,用浓度因素代替标准液进行检测,经精密度和回收试验,标准差为0.68,变异系数为0.49%,回收率均值为99%。获得满意结果。 Analysis of serum bilirubin plays a very important rule in the doagnosis,management and prognosis of hepatobiliary diseases.According to the recent survey of interlaboratory qualily control the bilirubin concentrations determined for one lot of bilirubin control serum were ten times in difference. The cause for such significant variation was surpposed to be the instability of bilirubin standards used.It was observed that bilirubin standards in 0.1mol/L sodium carbonate and 0.1mol/L sodium hydroxide decomposed at rate of 2% and 10% per hour and the half-life were 39 hours and 20 hours respectively. We observed that changes in the abssorbance of bilirubin standards stored in refrigerator in 20 consecutive days were significant and almost half of its content was decomposed.Tne bilirubin standard is very expensive apart from its instabitity.During our experience in the automatic biwavelength examination of serum bilirubin, we tried an alternative way to overcome the draw backs of bilirubin standard factor in the determinition of serum bilirubin.Tne confficient of variation at 1.2% (X=152.6μmol/L) and 4.4% (X=23.gμmol/L) were attained and the recoveries were 96% to 98% in the range from 88.9μmol/L to 140.2μmol/L. We found it a precise way to replace the use of bilirubin standards in routine analysis as well as economical.
