抗远缘链或菌OMZ176表面蛋白抗原（220kD）单克隆抗体的制备

Preparation of Monoclonal Antibody to the Cell wall Protein Antigen 220kD of Streptococcus Sobrinus OMZ 176

采用远缘链球菌ＯＭＺ１７６（ｄ血清型）全菌细胞作抗原，通过免疫ＢＡＬＢ／ｃ小鼠，细胞融合，杂交瘤细胞的筛选及亚克隆，最终获得抗远缘链球菌ＯＭＺ１７６表面蛋白抗原（２２０ｋＤ）的单克隆抗体（ＷＣ３Ａ６）。结果表明：ＭｃＡｂ的Ｉｇ及亚类鉴定为ＪｇＧ２ａ，ＥＬＩＳＡ效价１：８０００，Ｗｅｓｔｅｒｎｂｌｏｔｉｎｇ鉴定可见ＭｃＡｂ与２２０ｋＤ蛋白抗原发生特异性结合。与变形链球菌ＭＴ６Ｒ（Ｃ血清型）有弱的交叉反应（ＯＤ值＝０．２３）。

Bacterial cell suspension of streptococcus sobrinus OMZ 176(serotype d)was used to immunize BALB/c mice.Hybridomas were made by fusing the splenocytes of the mice with Sp2/0 myeloma cell.Hybrids were screened by testing the hybridoma culture supernatant(HCS)in ELISA.McAb(WC3A6)which reacted with S. sobrinus OMZ 176 surface protien antigen 220kD was obtained.The results indicated that the subclass of the antiboby was IgG2a,the HCS titer was 1: 8,000 and the specific reaction betweell McAb and OMZ 176 surface protein antigen 220kD was shown by western blotting.There was weak cross-reaction(OD=0.23)with streptococcns mutans MT6R(serotype c).
