摘要
用六种限制性内切酶BamHⅠ、EcoRⅠ、PstⅠ、BglⅠ、BglⅡ和SalⅠ对滑鼠蛇肝脏线粒体DNA(mtDNA)进行酶解。发现BglⅡ、PstⅠ、BamHⅠ、BglⅠ和EcoRⅠ在滑鼠蛇肝mtDNA上分别有1、2、3、3和4个切点。SalⅠ不能切割滑鼠蛇肝mtDNA。根据滑鼠蛇肝mtDNA的单酶、双酶完全酶解及部分酶解片段的数目和分子量,建立了滑鼠蛇肝mtDNA的限制酶图谱。
The mitochondrial DNA(mtDNA)from Ptyas mucosus liver was digested with six restriction endonucleases. A unique site for Bgl Ⅰ , 2 sits for Pst Ⅰ, 3 sites for BamH Ⅰ and Bgl Ⅱ , and 4 sites for EcoR Ⅰ were found on the mtDNA, however there was no site for Sal Ⅰ. The mtDNA was cleaved by double enzymes digestions and partial digestions with the above mentioned enzymes. The framents thus obtained and their molecular weights were estimated by agarose gel electro-phoresis.A restriction map was made according to the number and the size of those fragments obtained by complete, partial and double digestions.
