菌紫质、磷脂与水的相互作用:红外光谱与吸附重量研究

摘要 应用紫膜等温吸附及红外差光谱技术研究细菌紫膜中水与蛋白、磷脂的相互作用.观察到酰胺Ⅰ、Ⅱ、Ⅲ带在R<0.05及0.08<R<0.14内有两阶段的变化,前者更为显著;而磷脂部分的改变发生在0.08<R<0.12范围.紫膜的水合顺序是先蛋白质后磷脂.讨论了蛋白、磷脂;水三者的相互关系,膜结构整合性在膜功能中的作用. IR difference spectroscopy and thin film adsorption isotherm measurement have been used to study the interaction of protein and phospholipid with water in purple membrane. It has been observed that Amide I,II,III have two step change in R<0.05 and 0.08<R<0.14. The first step change is greater. The absorption bands of phospholipid show alteration in 0.08<R<0.12. The hydr-ation sequence occurs in such way that the protein is followed by phospholipid. The interelation between bacteriorhodopsin, phospholipid and water, and the significance of integrity of membrane in its function implementation have been discussed.
