摘要
用流式细胞仪、免疫细胞化学和免疫透射电镜方法,分析研究不同剂量γ线体外照射人外周血T淋巴细胞后,不同时间细胞表面CD3抗原的表达和分布。发现随照射剂量增加,CD3阳性细胞百分率减少,同一照射剂最随照后时间延长,CD3阳性细胞百分率缓慢增加。受照细胞胞浆内游离CD3随照射剂量增加而增多,并于浆内出现CD3-SPA-胶体金颗粒(未照射细胞只见于细胞表面。全未见于浆内)。CD3、CD4双阳性流式细胞仪分析的结果与CD3单阳性分析结果基本一致,同时发现CD3、CD4双阴性细胞随受照剂量增加而增多,随照后时间延长而减少。
Human T-cells were irradiated in vitro and the CD3 antigen expression and its distribution in the irradiated cells were determined with flow cytometry,immunocytochemistry and immunoelectromicroscopy in different time intervals after exposure.It was found that CD3 antigen expression was decreased on the cell surface but increased in the cytoplasm along with the increase of radifltion dosage,the percentage of CD3 positive cells was slowly increased and the free CD3 in the irradiated cells were increased alogn with the increase of radiation dosage;The analytic findings of double positive cells to CD3 and CD4 were essentially similar to those of single positive cells to CD3.It was also found that double negative cells to CD3 and CD4 were increased along with the increase of radiation dosage but decreased along the prolongation of exposure time.
