摘要
作者建立检测二氯乙酸钠血清浓度的紫外分光光度法,平均回收率为101.4%,日内变异为3.3%,日间变异为3.8%,最低检测浓度为1mg/L.应用本法研究二氯乙酸钠(iv,180mg/kg)在兔体内的药代动力学,主要药代动力学参数为:T1/2=0.67h,Vd=0.28L/kg,K=1.1h-1,Cl=0.31L/kg/h,AUC=605.1mg·h/L.
The authors determined serum sodium dichloroacetate(DCA-Na)content in rabbits using ultravio-let,spectrophotometry with a wavelength of 265 nm.The mean recovery rate was 101.4%,the intra-day and inter-day cv was 3.3% and 3.8%,respectively,and minimal detectable level was 1 mg/L.Pharmacokinetic study on DCA-Na was carried out in rabbits receiving a single intravenous injection of DCA-Na (180 mg/kg).The changes in serum DCA-Na concentration were best described by a 1-compartment open model using PKBP-N1 program.The main parameters obtained were as follows:T1/2 0.67 h,Vd0.28 L/kg,Cl0.31 L/ kg/h and AUC 605.1mg·h/L.
