摘要
作者研究了白细胞介素-1α(Interleukin-1α,IL-1α)对培养的牛脑微血管平滑肌细胞增殖的影响。在5~500pg·ml-1范围内,IL-1α可诱导培养的牛脑微血管平滑肌细胞增殖,当用50pg·ml-1IL-1α与该平滑肌细胞共孵育24h时,刺激作用最强,增殖率达29.1%。欧芹素乙、异欧芹素乙,6-(α,α-苯基乙酰哌嗪基苯基)-4,5-二氢-5-甲基-3(2H)哒嗪酮在浓度为10-6~10-4mol·L-1时,可剂量依赖性地拮抗IL-1α诱导的该平滑肌细胞增殖,药物浓度最高(10-4mol·L-1)时,抑制率值最大;6-(α,α-二苯基乙酰哌嗪基苯基)-4,5-二氢-5-甲基-3(2H)哒嗪酮,在低浓度时(10-6和10-5mol·L-1)可抑制IL-1α诱导的该平滑肌细胞增殖,药物浓度升高,抑制率也加大,而在高浓度时(10-4mol·L-1)抑制率下降。
The effects of Interleukin-1 alpha(IL-1α) on the proliferation of bovine cerebromicrovascular smooth cells(BCSMC) were investigated. At concentration from 5 to 500 pg·ml-1, IL-1α induced proliferation of cultured BCSMC. At 50 pg·ml-1, after 24 h incubation with BCSMC, stimulative effect of IL-1α reached maximum: 29.1%. At concentration from 10-6~10-4 mol· L-1, imperatorin(Imp), iso-imperatorin(Isi) and 6-(α,α-phenylacetylpiperazinyl) phenyl-5-methyl-4,5-dihydro-3(2H)-pyridazinone(PMDP) antagonized IL-1α induced proliferation of BCSMC in a dose-dependent manner. At 10-4 mol·L-1, inhibitory rates of Imp, Isi and PMDP were respectively 62.1±2.1%, ;48.3±5.2% and 46.5±13.1%. At 10-6 and 10-5 mol.·L-1,6-(α,α- diphenylacetylpipezinyl) phenyl-5-methyl-4,5-dihydro-3(2H)-pyridazinone(DMDP) inhibited IL-1α- induced proliferation of BCSMC, and their inhibitory rates were respectively 34.3±3.5% and 56.6± 9.8%. At 10-4 mol·L-1, inhibitory rate of DMDP reduced by 28.3±4.6%.
