摘要
采用两类单抗进行肺癌的放免显像,即抗小细胞肺癌单抗2F7和抗CEA单抗,2F7单单抗并经胃蛋白酶裂解,得其片段F^131I标记用Iodogen法,标记率80%。共对36例肺部病变作了放免显像,其中小细胞肺癌25例,非小细胞肺癌病变11例。在25例小细胞肺癌中,19例行^131I-2F7显像,结果17例阳性,2例阴性;另6例行^131I-2F7F显像,结果5例阳性,1例阴性。
Radioimmunoimaging(RII) in lung cancer was carried ort by using radiolabelled MoAb 2F7 and anti-CEA MoAbs(including C6,C16,C50 mixture)in 36 patients.2F7 is an antihuman small cell lung cancer(SCLC)MoAb.Based on 2F7 MoAb 2F7F(ab')2 was generated by pepsin digestion.2F7 2F7F(ab)2 and anti-CEA MoAbs were babelled with 131I by Iodogen.Thirty-six cases(30 males and 6 females,age from 35 to 70) formed the study population.All patients were pathologically proved.Out of 36 patients 25 were SCLC,and 11 were non-SCLC.Among 25 SCLC cases,19 were gamma scanned with 131I-2F7,in whom 17 cases were positive while 2 showed negative images;the other 6 cases were tested with 131I-2F7F(ab')2,in whom 5 cases were postive and 1 showed negative result.Eleven patients were imaged with 131I-CEA MoAb,in whom 5 were adenocarcinoma,4 squamous cell carcinoma and 2 inflammation.Four cases were positive and 1 was negative in 5 of adenocarcinoma.In 4 cases of squamous cell carcinoma 2 were positive and 2 were negative;2 cases of infection were negative.These results confirmed that image quality and sensitivity of 131I-2F7 and 131I-2F7F(ab')2 MoAb are better than those of 131I-ECA MoAbs.It may be due to no CEA antigen expression in part of lung cancer cells.
