摘要
本实验将带有人肿瘤坏死因子。基因(TNF-α基因)cDNA的重组表达质粒直接注入小鼠骨骼肌内,观察了外源基因在小鼠体内的表达情况。双抗体夹心法ELISA法显示,实验组小鼠肌注15d后血浆中TNF蛋白含量逐渐增高并达高峰,然后开始下降。RNAdotblot结果显示,肌注20d时实验组中ELISA阳性的小鼠,其肌注处肌肉组织有TNF-α基因表达。由此表明,直接将外源DNA导入骨骼肌细胞内是基因转移的又一新途径。
It was found that an injection of the recombinant TNFαcDNA plasmid directly into the skeletal muscle of mice resulted in a significant expression of TNF gene in the muscular cells. Human TNF protein could be detected in the mice 15 days after injection with ELISA and RNA dot blot. It is concluded that direct introduction of exogenous DNA into the cells of skeletal muscles will be a new route for gene transfer.
