摘要
为研究大鼠垂体黄体生成素的α和β亚基的生物合成及分泌,首先需要得到高纯度的促性腺细胞。继往,人们曾使用密度梯度离心法来分离不同的垂体细胞,但其细胞存活率和回收率都比较低。我们使用离心分选技术来处理经酶解的完整大鼠垂体细胞,虽细胞成活率及回收率明显提高,但并不能将促性腺细胞与促甲状腺素分泌细胞适当分开。然而应用阉割30天的雄性大鼠的垂体进行实验,经离心分选后,可得到相对纯净的促性腺细胞群,并可使之与促甲状腺素分泌细胞相对分开,离体培养条件下,加入促性腺激素释放激素,可以刺激较纯净的促性腺细胞分泌LH增加。因此我们认为用离心分选技术所得之相对纯净的促性腺细胞可以作为研究大鼠LH亚基合成及分泌的离体模型。
Since the alpha subunit is common to all pituitary glycoprotein hormones, it is desirable to have enriched population of gonadotropes for studying the regulation of alpha and beta subunits of rat luteinizing hormone. Centrifugal elutriation of enzymatically dispersed rat pituitary ceils obtained from intact male rats, did not adequately separate gonadotropes from thyrotropes. However, when pituitaries were obtained from the castrated rats 4 weeks after orchiectomy, elutriation led to reasonable separation of gonadotropes from thyrotropes.The enriched gonadotropes, maintained in monolayer culture, respond to gonadotropinreieasing hormone by an increase in luteinizing hormone secretion and can thus provide a useful in vitro model to study the synthesis and secretion of rat LH subsunits.
