摘要
目的:观察脂质体对日本脑炎病毒E糖蛋白的佐剂作用;方法:利用改进的振荡法,使亲和层析提取的日本脑炎病毒E糖蛋白包裹于脂质体中,制备出脂质体-E蛋白结合物,并分组免疫BALB/c小鼠;通过ELISA,血凝抑制试验(HI)和中和试验(NT)等指标观察.结果:脂质-E蛋白免疫组的特异性抗体水平明显高于游离E蛋白组;在两批病毒攻击保护性试验中,前者保护率分别为50%和57%,后者分别为38%和30%.结论:脂质体对日本脑炎病毒E糖蛋白有明显的佐剂作用.
Objective:In order to evaluate the adjuvantity of liposomes. Methods:Japanese Encephalitis virus(JEV) E-glucoprotein purified by affinity chromatography was encapsulated in the liposomes by hand shak-ing.Three groups of BALB/c mice were immunized peritoneally with JEV-E glucoprotein entrapped in lipo-somes(LE ),free E(FE glucoprotein and“ empty”liposomes(EL)respectively.The specific antibody was detected by ELISA,hemagglutination inhibition (HI)and neutralization test(NT)dynamically and the pro-tection rate of virus attack was observed.Results:The results show that the specific antibody level and pro-tection rate of the LE group are higher than those of the FE,In two batches of virus attack, the protection rates of the free E-protein group are 38%and 30%and those of the entrapped E-protein group are 50%and 57%. Conclusion:The study suggests that liposomes can promote the antigenicity of JEV E-glucoprotein.
