胃癌耐药细胞株耐药谱的体外实验 被引量:13

In vitro study of anti-cancer agents resistant patterns of resistant hu-man gastric cancer cell lines

摘要 目的:观察胃癌细胞与抗肿瘤药作用后对不同药物的耐受情况.方法:胃癌细胞系SGC7901在体外分别与长春新碱(VCR),5氟尿嘧啶(5-Fu),阿霉素(ADM)及氨甲喋呤(MTX)作用,获得SGC7901/VCR,SGC7901/ADM,SGC7901/5-Fu及SGC7901/MTX4个耐药细胞株.体外细胞毒性实验观察它们对VCR,ADM,5-Fu,MTX,顺铂(CDDP)以及丝裂霉素以C(MMC)的敏感性.结果:4株细胞对VCR,MTX,5-Fu及MMC均有耐药,耐药指数为2.9~1583.SGC7901/VCR和SGC7901/ADM对ADM分别呈1.6倍和101倍耐药.SGC7901/ADM和SGC7901/5-Fu对CDDP呈6.3和2.5倍耐药.结论:4株胃癌细胞均产生了多药抗药性(Multidrugresistance,MDR),药物引起的耐受可分为单向性交叉耐药及双向性交叉耐药两种类型. bjective:The resistant patterns of acquied resistant of gastric cancer cell lines to anticancer drugs were investigated.Methods:Four resistant sublines,SGC7901/VCR,SGC7901/5-Fu,SGC7901/ADM,SGC7901/MTX were established with gastric cancer SGC790l cell separately selected by vincristine(VCR),5-fluorouracil(5-Fu),adriamycin(ADM)andmethotrexate(MTX)in vitro.The susceptibility of sublines to VCR,ADM,5-Fu,MTX,cisplatin(CDDP)and mitomycin(MMC)were studied using MTT methods.Re-sults:Four resistant sublines were cross resistant to VCR , MTX , 5-Fu and MMC, resistant index varying from 2.9 to 1583.SGC7901/VCR and SGC7901/ADM cells were resistant to ADM by 1.5 and 101-fold re-spectively while SGC7901 /ADM and SGC7901/5-Fu resistant to CDDP by 6.3 and 2.5-fold.Conclusion:Four sublines are all multidrug resistance(MDR).Anticancer drugs can cause one-way cross resistance and two-waycross resistance.
