摘要
目的:为了研究人胃癌组织中癌基因及其调控序列是否存在结构上的改变,首先需要分离并获得所需研究的基因.方法:用人胃癌异常高表达基因cDNA片段(hSCG-3)和c-myc基因为探针,经同位素标记,对所构建的人胃癌组织基因组文库进行二轮噬菌斑原位杂交筛选.结果:从该基因组文库中分别筛选得到2个和6个阳性克隆.结论:为进一步探讨胃癌发生的机理,以及对这二个基因进行亚克隆和序列分析奠定了基础.
objective:To detect the structural mutation of the oncogene and its regulator gene in humanstomach cancer,the desired genes were separated and obtained in advance.Methods:A genomic library con-structed from human primary gastric cancer tissue was screened by in situ hybridization of bacteriophageplaques with two kinds of radiolabeled probes:One is an abnormally over-expressed gene(a cDNA fragment,named hSCG-3)and the other c-myc gene,Results:Finally 2 and 6 positive clones were obtained with thetwo probes,respectively,after two rounds of plaque screening,Conclusions:The subcloning and sequencingof the two genes will be further investigated.The result will provide further information about the mecha-nism of oncogenesis of stomach cancer.
