摘要
目的:建立大鼠脑脓肿模型,探讨脓肿周围组织ATP酶活性改变与脑水肿的关系.方法:大鼠右额叶定向注入细菌悬液(1μl,含金黄色葡萄球菌104个),接种后1~19d测定脓肿周围脑组织ATP酶活性及水含量.结果:大鼠接种细菌后均在右额叶内形成脓肿,其病理过程与临床情况相似;脓肿周围组织ATP酶活性在早期(7d内)持续性降低,而水含量则进行性升高.脑水肿与组织Na+K+-ATP酶(r=-0.865),Ca2+ATP酶(r=-0.881)活性改变之间呈高度负相关(P<0.01).结论:大鼠脑内特定部位定量种植细菌可稳定复制脑脓肿模型,脓肿周围组织ATP酶活性抑制与脑水肿发生发展密切相关.
Objective:To produce a brain abscess model in rats and to determine the changes of ATPase activ-ity in the tissues around the abscess and its relevance to brain edema.Methods:Using stereotactic technique,1 μl of bacterial suspended saline containing 106 Staphylococcus aureus was injected into the white matter ofright frontal lobe of rats to produce brain abscess.The ATPase activity and water content were measured ev-ery other days from 1st to 19th day after bacterial inoculation.Results:After injection of the bacteria,eachrat had developed a brain abscess in the white matter of right frontal lobe and the pathological process of thisbrain abscess was similar to that seen in clinical cases.ATPase activity was decreasing while water contentincreasing progressively in the surrounding brain tissues within the first 7 days after bacterial injection.Therewas a significant negative correlation between brain water content and the activities of Na+-K+-ATPase(r=-0.865,P<0.01),and Ca2+-ATPase(r=-0.881.P<0.01).Conclusions:The results indicate that in-fusion of a certain number of bacteria into a specific position of the brain could produce cerebral abscess con-sistently in rats.The inhibition of ATPase activity in the tissues around the brain abscess may be an impor-tant factor in the development of brain edema.
