免疫抑制酸性蛋白在BBN诱导小鼠膀胱癌变组织中表达的动态观察

摘要 作者用0.5g/LN-butyl-N-(4-hydroxybutyl)nitrosamine(BBN)诱导ICR小鼠膀胱癌变,10周后又用0.4g/L的丝裂霉素行膀胱灌注,在光镜及电镜下观察膀胱移行上皮细胞转化与转归的变化。然后用与长臂活化生物素结合的抗免疫抑制酸性蛋白(IAP)单抗及双ABC免疫组化法研究IAP在小鼠膀胱癌变组织中的动态表达,结果:正常小鼠膀胱移行上皮染色阴性,癌变的膀胱上皮染色阳性,并随癌肿的抑制而消退。 0.5 g/L N-butyl-N-(4-hydroxybutyl)nitrosamine(BBN)was used to induce bladder tumor inICR mice, followed by instillation of 0.4g/L Mitomycin-C(MMC)into mice bladder 10 weeks later。 Theprogression and regression of epithelial transitional cells in mice bladder tumor were observed with light andeleetron microscopes, In addition, the changes of the immunosuppressive acidic protein (IAP)expressions inthe mice bladder tumor tissue were also studied using biotinized MI2(conjugated with N-hydroxy-succinimido-6-biotinylamido hexanoate, BCNHS ) and double ABC immunoperoxidase histochemical stain assay。 The re-sults showed that the normal epithelium of mice bladder was negative , while the tumor growing epithelium ofmice bladder was positive and subsided gradually with the administration of MMC.
