摘要
电镜标本常规包埋过程比较复杂、费时,一般需60h。本实验采用高温快速包埋法。它是根据物质受热后分子运动加速,包埋介质粘稠度降低,缩短了浸透时间,减少了实验步骤。整个包埋过程仅用8h,而超微结构观察与常规方法比较没有明显差别,大大提高了工作效率。
The conventional embedding procedure for
electron microscopic samples is complicated and time-con-suming. Usually, it takes 60 hours.A
rapid high-temperature embedding method described here is basedon the acceleration of
molecular movement in the material and decline of viscosity of embedding medium athigh
temperature. The immersion time was shortened and a few steps were omitted in the new
method.Only 8 hours did it take to finish the whole embedding procedure, Moreover,the
ultrastructure had no ob-vious difference from that of the samples prepared by the conventional
method.Therefore,this method cangreatly elevate the working efficiency.
