摘要
羊促黄体生成素(oLH)的β亚基(oLH_β)是一种颇有潜力的抗生育疫苗。为筛选oLH_β的抗原决定簇(epitopes),开发由hCG_β-CTP 37和oLH_β最佳片段组成的复合疫苗,制备高纯度的oLH是必要前提。本文报道用抗oLH_β-IgG免疫亲和层析柱纯化oLH的新方法,得到了生物学活性相当于3,050·hCG IU/mg的oLH精制品,其电泳图谱显示高纯度的单一色带。本法具有纯度高、重复性好、操作简单的优点。
Some previous studies suggested that oLH beta subunit (oLH_β) is a potential vaccine for antifertility. In order to develop a mixed vaccine which consists of hCG-β-CTP-37 segment and the screened epitopes of oLH_β, the preparation of highly purified oLH is an essential prerequisite. A new methodology for preparation of ovine LH with high purity by anti-oLH_β-IgG immuno- affinity chromatography has been reported. The biological activity of oLH is corresponding to 3,050 hCG IU/mg, exhibiting a single band in PAGE. Its advantages are: high purity, good reproducibility and convenient operation.
