摘要
本工作采用离体孵育大鼠颗粒细胞的方法,观察了肾上腺素和去甲肾上腺素对颗粒细胞雌二醇生 成的影响。为进一步探讨其作用机制,实验还观察了β-肾上腺素能受体阻断剂心得安、外源性cAMP和 腺苷酸环化酶激动剂(Forskolin)对肾上腺素和去甲肾上腺素诱导颗粒细胞雌二醇生成的影响。结果 发现,肾上腺素和去甲肾上腺素可明显促进离体大鼠颗粒细胞雌二醇的生成,此作用可被心得安阻断, 外源性cAMP和Forskolin可促进肾上腺素和去甲肾上腺素诱导的颗粒细胞雌二醇生成。表明肾上腺素 和去甲肾上腺素促进大鼠颗粒细胞雌二醇生成的作用是通过特异地结合到颗粒细胞膜上β-受体,且可 能与激活细胞内腺苷酸环化酶-cAMP系统有关。
In Order to provide more direct evidences for that the catecholamine may play the role of regulation of ovarian endocrine function, an investigation on the effect of E and NE on E2 production using rat granulosa cells in vitro was carried out. Granulosa cells prepared from immature female rats after 48 h pretreatment with PMSG (100U/rat) were preincubated for 1h and then treated with or without E, NE or other agents alone, or in combination for a further 2 h, medium E2 was meadured by RIA. The results showed that both E (0. 54 and 1. 6mmol/L, respectively) and NE (0. 59 and 1. 8 mmol/L, respectively ) significantly stimulated E2 production by rat granulosa cells during the 2 h incubation. The β-adrenergic antagonist, propranolol(1mmol/L),inhibited E2 production which was induced by E/NE. In addition, exogenous bd cAMP (1mmol/L) and cAMP generating agent, forskolin (1mmol/L) enhanced both basal and E/NE induced E2 tion. The results indicate that E/NE stimulates E2 production by rat granulosa cells in vitro, and its action may be mediated by β-adrenergic receptor system and adenylate cyclase-cAMP system.
