摘要
应用自制硅胶板的薄层色谱法分离了红细胞膜上四种主要磷脂。将分离后的每种磷脂斑点硅胶涂层刮下,不经萃取,直接在无水甲醇-苯-乙酰氯溶液中进行转移甲基化,然后用毛细管气相色谱分离测定其脂肪酸组成和含量。上述方法已成功地用于先天愚型病人和正常人红细胞膜上各种磷脂中脂肪酸轮廓分析,获得有意义的结果。
the determination of fatty acids in various phospholipid was carried out by capillary gas chromatography as their methyl ester. The neutral lipids, phosphatidyl choline (PC), phosphatidyl ethanolamine (PE), phosphatidyl serine (PS) and sphingomyelin (SM) in the erythrocyte membrane of normal person and patient with mongolism were separated by TLC using home made plate. The separated band of the phospholipid was scraped and methylesterified in methanol-benzene with acetyl chloride as catalyst. The fatty acid esters from various phospholipids were separated on a bonded OV-225 fused silica capillary column and their contents were determined using 17:0 methyl ester as internal standard. The com-parison of the composition of fatty acids in various phospholipids between from normal person and from patient with mongolism was made. The results show that the content of satu-rated fatty acid 16:0 in the PE and PC from patient with mongolism is higher than that from normal person (P<0.001), while content of unsaturated fatty acid, 20:4 in PE, 18:1 and 18:2 in PC from patient with mongolism is lower than that from normal person (P<0.001)
