增强化学发光酶免疫分析法测定血清中的铁蛋白

Enhanced Chemiluminescence Enzyme Immunoassay for the Determination of Ferritin in Serum

本文采用对碘苯酚增强的Luminol(鲁米诺)-H_2O_2-HRP(辣根过氧化物酶)化学发光反应体系作为免疫分析的最终检测手段,建立了一种新的铁蛋白(Ferritin)的免疫分析方法,与酶联免疫分析(ELISA)法相比,该方法具有灵敏度高,线性范围宽等优点。方法的相对标准偏差为3.7％(1:896000倍稀释的铁蛋白抗体(anti—ferritin)-HRP,n=11),标准曲线线性范围为0.12～2000ng/ml,用该方法对血清中的铁蛋白进行了测定,其结果与ELISA法所得结果一致。

A new immunoassay method for the determination of ferritin which is used as end-point monitoring system in para-iodophenol enhanced luminol-H2O4-horseradish peroxidase (HRP) chemiluminescence reaction was developed. Compared with conventional enzyme -linked immunosorbent assay (ELISA) , this method offers advantages of high sensitivity, wide linear range and good precision (relative standard deviation for 1 : 896000 fold diluted anti-ferritin-HRP is 3. 7% ,n = 11). The linear range of standard curve is 0.12-2000 ng/ml. The method has been used for the determination of ferritin in serum, and the results is conformable with those obtained ELISA.
