Determination of Chlorophyll a and Chlorophyllb in Vegetable Leaves by First Derivative Spectropho-tometry

本文提出用一阶异数分光光度法同时测定蔬菜叶片中叶绿素ａ和叶绿素ｂ的含量。讨论了同时存在于蔬菜叶片中的类胡萝卜素对两者测定的影响，并提出用适当浓度的类胡萝卜素丙酮溶液作为参比溶液，用以消除类胡萝卜素对叶绿素测定的影响。

A first derivative spectrophotometric method ispresented for the determination of chlorophyll a andchlorophyll b in green vegetables. Chlorophyll a is de-termined at 671nm using an acetone solution as thereference solution. Chlorophyll b is determined at467nm using a carotenoid solutiOn as the reference so-lution, The linear correlation coefficients of standardcurves are equal to 0.9999747 and 0.9999859 forchlorophyll a and chlorophyll b respectively. The re-coveries are between 100.5%and 101.8%for chloro-phyll a.
