摘要
用离子选择电极电位法研究了银和铅与人白蛋白(HSA)的配合平衡。明确证实银和铅都与HSA形成了单核配合物MA和MA2.Ag-HSA配合物的稳定性积β(11)=4.4×107,β(12)=1.0×10(12);Pb-HSA配合物的β(11)=3.1×105、β(12)=2.0×109。根据HSA与人白蛋白羊抗血清(GAHA)的免疫反应,用银和铅分别标记HSA时,测定GAHA(效价1:40)的线性范围是0.2~2.0μL和0.4~2.8μL;而用银和铅分别标记GAHA时,测定HSA的线性范围是0~2.9×10(-8)mol/L和0~7.3×10(-7)mol/L。
The complex equilibria of Ag-human albumin (HSA) and Pb-HSA were studied by ion-selective electrodes.It was clearly proved that both Ag and Pb with HSA formed mononuclear complexes MA and MA2 respectively.The stability products of Ag-HSA complexes are β(11)=4.4×107 and β(12)=1.0×10(12);and that of Pb-HSA complexes are β(11)=3.1×105,β(12)=2.0×109.According to the immunochemical reaction of HSA with albumin antiserum (GAHA),the linear range of determing GAHA(titer 1 : 40)is respectively 0.2-2.0μL and 0.4-2.8μL when HSA is labelled by Ag and Pb; moreover the linear range of determining HSA is respectively 0-2. 9×10(-8)mol/L and 0-7.3×10(-7)mol/L when GAHA is labelled by Ag and Pb.
