煤焦沥青与膜的脂质过氧化 被引量:4

摘要 本文研究了煤焦沥青(CTP)在体内及体外条件下对膜脂质过氧化(LPO)的影响。在体内试验中,48只Wistar大鼠分为二组。实验组气管注入10mgCTP粉,对照组气管注入10mg碳粉。一周后开始剖杀检查,8周内分别观察大鼠肺、肝、肾的胞液、线粒体、溶酶体及微粒体的LPO改变以及SOD、CAT和GSH-PX活性。结果未发现两组有明显差别。在体外试验中,CTP、CCl4、BaP及DMBA在相同条件下分别与微粒体—NADPH体系反应。结果CTP呈现强烈的抑制LPO作用,CTP在体内试验中所以不能显示对LPO的抑制,可能是由于CTP在体液中的溶解度太低,达不到抑制LPO所必需的浓度。至于CTP对LPO的增强作用则无论在体内或体外试验中都得不到证实。 Anin vivo and in vitro assay on the influence of coal tar pitch(CTP) on membrane lipid peroxidation( LPO) is reported.In the in vivo assay 48 Wistar rats were divided into 2 groups. 10mg CTP powder suspended in 0.2ml normal saline solution were injected intratracheally in each rat for the experimental group and 10mg carbon powder in each rat for the control.One week later,LPO and SOD,CAT and GSH -PX activities of the membrane components including cytosols, mitochondria, lysosomes and microsomes of lungs, liver and kidneys were examined once two weeks until the 8th weekend. No significant differences in LPO values and enzyme activities between the two groups were observed. In the in vitro assay CTP. CCl4, BaP and DMBA were tested separately with the mocrosome-NADPH system at the same experimental condition.The results showed that CTP exerted a strong inhibitory effect on LPO.The reason that the inhibitory effect of CTP did not appear in the in vivo assay might be the insolubility of CTP in the body fluid which did not permit CTP reaching the necessary concentration for exerting the inhibitory activity. No increase of LPO was observed either in vivo or in vitro assay
