摘要 通过使用诊断性底物,多克隆抗体和血液自动分析仪,我们检查了乙酰肼对大鼠肝微粒体细胞色素P450、P450亚型及血液生化学指标的影响。结果提示:乙酰肼可引起大鼠肝脏微粒体苯胺羟化酶活力增加,但苄甲异丙胺N-脱甲基活力降低;P450亚型2E增加,但P450总量减少;血清GOT,GPT的量也减少。本研究证实:乙酰肼选择性诱导P4502E,同时抑制肝微粒体其它P450亚型。 The influence of acetylhydrazine(AHZ) on the hepatic microsomal cytochrome P450 and serum biochemistry were investigated in male rats. Changes in specific cytochrome P450 protein were monitored by using diagnostic substrates and immunologically polyclonal antibody.The activity of aniline hydroxylase,when expressed in terms of nmol. mg-1 microsomal protein, was increased by the treatment with AHZ.In contrast, the activity of benzphetamine N-demethylase significantly decreased.Immunoblot analysis showed that P450 2E was increased in AHZ treated rats, while the total microsomal cytochrome P450 was decreased.The serum level of GOT, GPT was also lower than that of control. These findings ihdicated that AHZ selectively induced microsomal cytochrome P450 2E while it repressed other microsomal P450 isozymes.
