僧帽牡蛎三倍体的研究 被引量:25

TRIPLOIDIZATION OF OYSTER OSTREA CUCULLATA WITH CTYOCHALASIN B AND TEMPERATURE SHOCK

摘要 报道了用细胞松驰素B、高温休克和低温休克方法诱导憎帽牡蛎三倍体的实验结果.在25.0~26.0℃条件下,进行不同浓度梯度的细胞松驰素B(0.1、0.5、1.0和1.5mg/L)和不同的处理时间的诱导实验.结果表明,在受精后18min用1.5mg/L细胞松驰素B处理受精卵15min,三倍体诱导率最高,达87.5%.用高温休克方法,在受精后18min分别于32.0、35.0、37.0和39.0℃处理受精卵10min,三倍体诱导率分别为4.1%、37.0%、55.2%和42.5%.低温休克方法,在受精后22min于0~1.0℃的2.0℃分别处理受精卵10min,三倍体诱导率分别为36.7%和23.5%.实验性育苗结果显示实验组的幼虫和稚贝的生长速度明显比对照组快.还就细胞松驰素B和温度休克诱导三倍体的机制及对僧帽牡蛎胚胎及早期幼虫发育的影响进行了讨论. This paper is concerned in triploidy induced in oyster, Ostrea cucullata by cy-tochalasin B(C. B) and temperature shock. Triploidy induction trials were carried out using C. B treatment at four concentration 0. 1,0. 5,1. 0 and 1. 5 mg/L for different treatment duration. The highest frequency of triploid was 87. 5% obtained when treatment was conducted at 1, 5mg/ L C. B for 15 min at 18 min post-insemination. Heat shock was applied to fertilized eggs for 10 min at 18 min post-insemination at four temperature 32. 0,35. 0,37. 0 and 39. 0#; and the frequency of triploid was 4. 1%, 37. 0%, 55. 2% and 42. 5% respectively. Cold shock was applied to fertilized eggs for 10 min at 0-1. 0# and 2. 0# at 22 min post-insemination and the frequency of triploid was 36. 7% and 23. 5% respecively. Daily growth of larvae and spat produced by inhibiting the first polar body with heat shock was higher that the control group. The mechanism and effect of C. B treatment and temperature shock on embryonic and early larval development of polyploid inducement wete discussed.
