杂种犬11只,快速股动脉放血使其平均动脉压(MAP)达6.0kPa左右,维持90min后输回全部血液,观察240min。维拉帕米(Verapamil,Ver)组于休克30min时静脉输注Ver150μg/kg,15min内输毕。实验结果发现,Ver治疗后,MAP高于休克组,在休克240min时两组差异有显著性。Ver治疗还可以降低胰腺组织黄嘌呤氧化酶(XO)活性,减少脂质过氧化产物丙二醛(MDA)含量,提高超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-px)活性。并能减轻胰腺组织的线粒体和内质网损伤。提示Ver对失血性休克犬的胰腺具有保护作用。
This study was designed to investigate the benefcial effect of administration of verapamil on pancreatic ultrastructure and lipid peroxidation during the hemorrhagic shock in dogs.Anesthetized dogs were rapidly bled to a mean arterial pressure(MAP) of 6. 0 kPa and maitained at that level for 90 min,followed by resuctation with all shed blood. At 30 min into hemorrhagic shock dogs verapamil(10μg/(kg.min n=6) or same volume of saline(3 ml/kg, n=5) was infused intervenously over 15 min. At the end of experiment(240 min after hemorrhage and resuscitation) the pancreas samples were obtained and analyzed. The results showed that verapemil treatment significantly enhanced MAP recovery, ameliorated pancreas untrastracture damage, with little malonyldialdehyde, low xanthine oxidase activity and high activity of Mn-SOD, Cu-ZnSOD and GSH-Px as compared with those of control dogs. The date suggests that verapamil has a benificial effect on pancreas of dogs with hemorrhagic shock.
