摘要
分别选用冰冻切片水浴、睾丸整体水浴和特异性底物等酶孵育方法显示*丸LDH-X活性。结果表明,冰冻切片水浴和睾丸整体水浴后酶反应产物扩散严重,并可使曲细精管结构受到一定程度的损害。特异性底物方法无上述缺陷、LDH-X定位良好,不失为目前显示睾丸LDH-X活性的较好方法。
Lactate dehydrogenase isoenzyme
X(LDH-X)activities of testis were demonstrated insitu by using cryostat section bathing,whole
testis bathing and specific substrate alternately.The results showed that the reaction products
of cryostat section diffused seriously afterbathing in 65℃ for 30min;the structure of
seminiferous tubules was damaged after wholetestis bathing and enzymatic reaction products
were scattered;LDH-X could be shown wellby using specific substrate of LDH-X.The results
suguest that using specific substrate is anavailable method for demonstrating LDH-X
histochemically in situ.
