摘要
采用PCR/SSO方法分析了67例湖南籍汉族正常人中HLA-DR,DQ,DP位点共105个等位基因的分布情况。在67例检出的72个等位基因中,其部份频率明显不同于美国白人、黑人和巴西混血人种。提示湖南汉族人与美国白人、黑人和巴西混血人种间存在明显的种族差异。
his paper reports that the DNA polymorphism analysis of HLA DR,DQ,DP in 67 cas-es
of Hunan Hans was performed by using the method of PCR to amplify the second exon
ofDRBl,DRB3,DRB4,DRB5,DQAl,DQB1,DPAl,DPBl,and of dot blot hybridization withradio-labeled
sequence-specific oligonucleotide probes. In order to resolve the cross hybridiza-tion of
oligonucleotide probes,the generic amplification and group specific amlification ofPCR primers
were performed respectively. The 55 DR alleles,9 DQAl alleles,17 DQBl alle-les,4 DPAl
alleles,and l9 DPBl alleles were determined.The results showed that the fre- quencies of
HLA-DR,DQ,DP alleles of Hunan Hans are significantly different from those ofCaucasian and
Blacks. The results of allele frequencies of DR,DQ,DP may be reference mate-rials in
association with diseases in Hunan province.
