摘要
自制微量进样装置,建立微量进样火焰原子吸收光度法。探索该法线性范围在0~6.0μg/ml,回收率96.4~101.3%,变异系数(精密度)为1.85~3.88%,最低检测限为0.02μg/ml。自大鼠双耳蜗吸取外淋巴液,混合后加 La 溶液稀释,再用原子吸收分光光度计微量进样,测定实验性佝偻病大鼠耳蜗的外淋巴钙浓度,获得满意效果。证明该方法具可行性,且简单易制.
A method for determining perilymph calcium in the rats' cochlea by microsampling atomic absorption spectroscopy was studied. Thirty-six Sprague-Dawley white rats suffering from rickets were killed after anaesthetization by sodium pentobarbital. A 5μl micro-injector was used to collect μl of perilymphatic fluid under an operating microscope. Each sample was diluted with 100 μl of 0.5% La solution. The perilymphatic calcium was then determined. The instrument conditions were as follows: lamp current 10mA, wave-length 422. 7nm, slit width 0. 5nm,air flow 13.51/min,acetylene flow 2.801/min,and sampling volume 50μl. The linear range,the precision and the recovery of the method were 0~6μg/ml, 1.85~3.88%, and 96.4,101.3% respectively. The method was used for the study of the perilymphatic cal- cium concentration of rickety rats and a satisfactory result was achieved.
