摘要
以“HoMei”和“东京笋”两个白菜品种为材料,探讨了植株栽培的环境条件对白菜小孢子培养的胚状体发生和植株再生的影响。结果表明,对胚状体发生能力较强的HoMei品种,环境条件对小孢子培养有一定的影响,生长在长日照(14~18h)和15~20℃温度条件下的植株,胚状体发生数及植株再生率显著高于短日照(12h)、较高温度下(25℃)生长的植株。对胚状体发生能力较弱的东京笋品种,环境条件对小孢子培养影响显著,生长在14h日照条件下的植株,产生的胚状体数显著高于其他条件下栽培的植株,温度条件以15℃的栽培条件为最好。
Two cultivars (Ho Mei,Tokyotakenoko)were used and the influence of growth conditions of donor plants on microspore culture in Chinese cabbage was investigated.The embryo yield of 'Ho Mei' and the proportion of torpedo and cotyledonary embryos were much higher than those of 'Tokyotakenoko'. When the donor plants of 'Ho Mei' were cultivated under long day length conditions (14, 16, 18hr),the embryo yields, the number of embryos which developed shoots longer than 2 cm and the percentage of plantlert regeneration were much higher than those cultivated under the short day length (12hr).But there were no significant differences between the donor plants cultivated under 14, 16, 18hr conditions. In the temperature condition study,the highest embryo yield, was obtained from the plants cultivated under the 20℃ growth condition. The results of 'Tokyotakenoko' were different from those of 'Ho Mei'. When the donor plants of 'Tokyotakenoko'cultivated under the 14hr condition, the embryo yield, the number of embryos which developed shoots longer than 2 cm and the percentage of plantlet regeneration were highest.With the donor plants under the short day length condition (12hr ),very few embryos were found and almost no plantlet was formed. The highest embryo yield was obtained from the plants under 15℃ and 14hr day length growth conditions
