摘要 以体外培养的猪主动脉内皮细胞(EC)为模型研究I1oprost对EC抗血栓功能的影响。用放免法、发色底物法测定培养液中6-酮-PGF1a(6-keto-PGF1a),血栓素B2(TXB2)含量,纤溶酶原激活物(PA),纤溶酶原激活物抑制物(PAI)活性及EC环磷酸腺苷(cAMP)含量。结果表明:I1oprost可以使EC生成6-keto-PGF1a,cAMP含量显著增多,TXB2含量显著减少,并使PA活性升高,PAI活性降低。提示:I1oprost通过促进抗凝、纤溶作用,能够增加EC的抗血栓功能。 y using cultured porcine aortic endothelial cells(EC)as an experimental mo-del in vitro 6-keto-pGF1α,TXB2 content, plasminogen activator (PA), plasmino-gen activator inhibitor(PAI)activity in the supernatant and cyclic AMP level in ECwere measured with RIA and the method of chromogenic substrates.The results showed: after administration of I1oprost 6-keto-pGF1α,cyclic AMPcontent and PA activity ,were significantly higher,TXB2 content, PAI activitywere remarkably lower than control group. These effects were dose dependent. These findings indicated that I1oprost might act as a prospective drug for anti-thrombosis through promoting anticoagulation and fibrinolysis function, increasingantithrombus action of EC.
