激光辐照葡萄愈伤组织的过氧化物酶同工酶分析

摘要 利用法国产DATACHROM—5000型染料脉冲激光器处理葡萄“胜利”品种的愈伤组织,用聚丙烯酰胺凝胶电泳法(PAGE)分离过氧化物酶同工酶,研究了激光辐照对葡萄生长发育和过氧化物酶同工酶表达的影响,发现:适当剂量的激光辐照能使葡萄愈伤组织明显提前分化;激光能够影响葡萄过氧化物酶同工酶基因的调控,适当剂量的激光能够促进该基因的表达;激光处理的愈伤组织在不同的生长发育时期与对照相比,过氧化物酶同工酶在数目和活性上有明显增加。 Grapevine callus of Vitis Vinifera L. CV. Triumph were irradiated by using DATACHROM 5000 type laser machine made in France. Peroxidase isoenzymes were separated by polyacrylamide gel elec-trophorosis. We found that:callus irradiated with appropriate dose of laser could differentiate ahead of timeobviously;laser irradiation could affect the regulation of peroxidase gene and appropriate dose of laser couldpromote the expression of peroxidase gene;compared with the control group,the kinds and activity of perox-idase isoenzymes of grapevine callus treated with laser were taised Obviously.
