油菜芜青花叶病毒鉴定、提纯及血清学 被引量:3

摘要 通过寄主范围、粒子形态、血清学测定,鉴定出南京油菜上的两个分离物为芜青花叶病毒(TuMV),T-02分离物表现为黄斑花叶型症状,称为花叶型株系(TuMV-jm);T-05为枯斑系统坏死症状,称为坏死型株系(TuMV-jn)。提纯研究表明,A_(260)/A_(280)为1.22;A_(max)/A_(min)为1.12;粒子长度为720—800nm。SDS聚丙烯酸胺凝胶电泳测定外壳蛋白亚基的分子量为27.5kd。还研究了外壳蛋白17种氨基酸的含量及比例关系。以提纯TuMV-jn制备抗血清,研究其免疫分析方法及其与马铃薯Y病毒组某些病毒的血清学关系。 According to studies on the host range,particle property and serology of 2 isolates,T-02 and T-05,from rape in Naning,they were identified as turnip mosaic virus (TuMV).T-02 was suggested to be amosaic strain(TuMV-jm)showing yellow mosaic symptoms and T-05 a necrotic strainTuMV-jn)showing system necrosis in most cultivars of rape.The Purified TuMV showed the following characteristics:A(260)/A(280)=1.22,Amax/Amin=1.12 and particle length 720 800 nm.The yield of purified virus was 1 4.0 mg/100g infected leaves.On the basis of SDS-PAGE of dissociated capsid protein of TuMV,each subunithad a molecular weight of approximately 27.5 kd.Comparison of amino acid composition of protein subunit was carried out.The antiserum prepared with purified TuMV-jn had titers of 1:2048 in micropreciptin test and 1:100000 in indirect ELISA.The 2 strains of TuMV are different from other members of the potyvirus group but the same between themselves in serology.
