热处理对猪胚耐低温性的影响

摘要 手术(或屠宰)采取猪囊胚(B)、扩张囊胚(EB)、孵出囊胚(HB),分别进行热处理(HS)、蛋白合成抑制剂(CH)和低温应激(CS)试验。结果,热处理与蛋白合成抑制剂对猪胚的生存率及发育率均无影响(P>0.05);而热处理对提高猪胚的耐低温性有显著效果。热处理组经低温应激后,24小时培养的EB和HB的存活细胞数分别较对照组提高35.5%-41.3%和51.1%,差异显著(P<0.05),且蛋白合成抑制剂具有抬抗热处理的作用。上述结果显示,热处理有可能诱发猪胚产生拈抗第二次应激的应激蛋白,这为进一步研究猪胚冷冻和改进猪胚冷冻前处理提供了依据和可能途径。 Porcine blastocysts (B), expanded blastocysts (EB)and hatched blastocysts (HB)were collected by operation or slaugter and exposed to heat shock (HS),cycloheximide (CH)and cold chock (CS).HS and CH did not affect the suvival and developed rate of porcine embryos (p> 0.05).Howers, HS could increase tolerance of the porcin embryos to low temperatures.For instance,the live cells of EB and HB in the HS group cultured for 24h after cold shock increased by 35%-41.3%and 51.5%(p<0.05), respectively,compared with the control group. It was also found that this effect could be inhibited by CH. It might be that the porcine embryo treated by HS produce sme antistress-proteins to the secondary stress,Therefore the experiment has provided a possible way for turther study to improve pretreatment of the embryos to be forzen.
