导向治疗对肝癌肿瘤浸润淋巴细胞体外生长动力学的影响

The Effect of Directive Therapy on the Expansion in Number and Killing Power of the Tumor Infiltrating Lymphocytes(TIL) Cultured in vitro for the Treatnent of Hepatic Carcinoma

对5份人肝癌标本的肿瘤浸润淋巴细胞（TIL）进行了体外培养，观察其生长动力学及杀肿瘤活性。结果显示：2份取自术前曾导向化疗且治疗有效的肝癌患者癌组织的TIL，分别于体外培养的第7、第8天即可见TIL细胞数有较明显增加，于第14、16天增殖达高峰，以后则增殖减慢。1份取自曾治但无效癌组织和另2份取自未治癌组织的TIL分别于体外培养的第18、19和16天方显示细胞数增加，于第30、33天增殖达高峰。所做的10次细胞毒性试验，有7次显示TIL对自体瘤细胞的杀伤率显著高于对肝癌细胞株的杀伤。

Tumor-infiltrating Lymphocytes (TIL) obtained from 5 human liver cancer cases were cultuned in vitro for investigating their numerical expansion and cytotoxicity. Among them, 2 tumors derived from patients who had effective direct-treatment, 1 tumor derived from patient who had been treated but bad no response, 2 tumors derived from patients who hadn't been treated. In result,the former 2 TIL cultures were successfully expanded in numter after culture for 7-8 days,and the time required for maximum lymphocyte expansion ranged from 14-16days. The other 3 TII culture growth became marked after 16, 18 and 19days and the maximum TIL expansion time ranged from 30 to 33days. Besides,in 7 of 10 cytoxicity assays TIL lysed the autologous tumor target to a greater degree than that lysed the liver cancer cell line- These results indicate that the preoperative direct-therapy can reduce the latent period of proliferative potentiality of TIL and they rnay still keep their specific cytotoxic effectto the tumor.
