草珊瑚细胞悬浮培养之研究 被引量:12

摘要 报道了草珊瑚(Sarcandraglabra)细胞悬浮培养的某些特性。细胞悬浮培养的摇床转速对草珊瑚细胞生长至关重要、培养物在100r/min速度下振荡培养几乎难以存活,最佳的摇床转速为40r/min(23.5h/d+80r/min(0.5h/d),其生长可增加5倍左右。细胞悬浮培养的接种量以2.0gFW/瓶合适,其培养物的月产率13.22gFW/瓶(或18.73gDW/1)。细胞生长的时间进程是,接种0-5d,细胞增殖十分缓慢,10-20d迅速生长,20d后基本停止生长。培养基的pH迅速下降时是细胞增殖缓慢的时期,当培养基pH值开始升高时,细胞增殖也显著加快。黄酮苷类物质的合成在0-20d迅速增加,20d左右达到高峰。 Suspension cell cultures of Sarcandra glabra have been established.The shaker speed in culturing suspension cells had an effect on the cellshaker speed 40 rpm(23.5 hr/day) with 80 rpm(0.5 hr/day) was considered the best for the cell growth.The yield of suspension cultures reached 13.22 g·FW/flask(or 18.73 g DW/1) per month with on inoculum of 2.0 g FW/flask.Time-course study showed that cell growth lagged in 0-5 days and enhanced greatly in 10-20 days and almost ceased after 20 days of cultivation.pH value was changing during the culture period.As the cell growth slow,pH value went down,and as the cell growth quick, pH value went up.When cells were cultured in liquid mediuin the content of flavonoid glycosides increased quickly and reached to the maximium about at the 20th day.
