摘要
生物化学分析表明,在北京鸭腔上囊发育过程中,淋巴细胞胞质内波形蛋白含量逐渐减少,而核内骨架成分变得丰富。IEF-SDS-PAGE双向电泳分析显示,216日胚腔上囊淋巴细胞核内骨架成分含20多种多肽,生后3周龄腔上囊淋巴细胞核内骨架成分含40多种多肽,并出现了分子量为35KD和37KD、等电点约为5.2和5.5的两个含量丰富的多肽。生后12周龄退化期的腔上囊的淋巴细胞核内骨架成分也趋于简单化,仅显示出10多种多肽。腔上囊淋巴细胞核纤层的主要成分是LaminB,而LaminA和C含量很少。腔上囊发育过程中淋巴细胞LaminB的含量呈渐增趋势。以上表明,在腔上囊发育过程中,淋巴细胞的中间纤维-核纤层-核内骨架体系蛋白质成分和含量有明显变化,其与腔上囊功能的关系值得深入探讨。
lectrophoretic patterns of SDS-PAGE and IEF-SDS-PAGE indicated that the changes of components of intermediate fiament-nuclear lamina-nuclear skeleton system in lymphocytes were obvious during the development of bursa of Fabricius. Immunoblot analysis demonstrated thht vi-mentin , a 55 KD protein , was the main component of intermediate filament in lymphocytes ofPeking duck. Quantitative analysis showed further that vimentin decreased during development of bursa of Fabricius. It was shown that about 24 dots 40 dots, and 15 dots could be clearly dis-tinguished in 2-D gel electrophoresis maps , which corresponded respectively to components of nu-clear skeltton in lymPhotytes of 26 day embryos and 3 weeks as well as 12 weeks after hatching.This result indicated that progressive changes in the protein composition of nuclear skeleton oc-curred during development of bursa of Fabricius. 2-D gel eltctrophoresis assay also showed that nuclear skeleton in lymphocytes taken from bursa of Fabricius of 3-week old duck contained two main polypeptides (35KD/PI5. 2 , 37KD/PI 5. 5) Molecular weight of lamins was about 67 KD. Lamin B, an acidic protein , was the main component of lymphocyte nuclear lamina and in-creased during development of bursa of Fabricius.
