摘要
分析了红细胞分化因子(EDF)对小鼠红白血病(MEL)细胞生长的抑制作用。当EDF剂量为0.12g/ml时,处理2d后的3H-TdR掺入率比对照组降低50%,第3d的细胞增殖抑制率达82%;在软琼脂上形成的集落数为对照组的0.52%。同时,处于细胞周期不同时相的细胞数量发生显著改变.G0加C1期细胞数比对照组增加55.66%,S期细胞数增加14.3%,而G2和M期细胞数则减少28.8%。此外,细胞电泳迁移率也减少到对照组的50%。以上结果说明,EDF对NIEL细胞的增殖有抑制作用。
The inhibitory effect of EDF on the growth of MEL cells in vitro has been analyzed in this study.We found that when the final concentration of EDF was 0.12μg/ml,the cell growth was inhibited,the 3H-TdR incorporation rates decreased by 50% at the 2nd day,the growth inhibitory rate of the cells was 82% at 3rd day,and the cells,colony formation ability on soft agar was 0.52% of the contrast.When the concentration of EDF was 0.10μg/ml,the flow cytometry experiment showed that the G0 and G1 phase cells were increased by 55.6%,the S phase cells were increased by 14.3%,but the G2 and M phase cells were decreased by 28.8%.In addition,the cell electrophoresis mobility rate also decreased to 50% of the original one after treated with EDF.All the results indicated that EDF possess inhibitory effect on the proliferation of the MEL cells.
