白细胞介素-1β对原代培养Leydig细胞白细胞介素-αmRNA基因表达的诱导作用 被引量:1

摘要 最近发现睾丸中存在IL-I样物质,但其来源不明,作者用改良的Klinefelter方法分离纯种大鼠睾丸Leydig细胞,培养于含有人重组IL-1β的牛血清的DEME/F12培养基中,根据Chomczyns-ki及Sacdchi方法,抽提全部细胞的RNA,最后用Northern印迹分析发现IL-β导致Leydig细胞中IL-1αmRNA表达增加,且呈剂量依赖性,脂多糖亦刺激IL-1αmRNA的表达,但佛波脂无此作用.结果表明Leydig细胞为IL-I的主要来源,它具有自分泌和旁分泌效应. Recently Interleukin-1(IL-1) like material has been reported to be present in the testis,however,the cel-luiar source and function remains unclear. In this study,we used modified method of Klinefelter to isolate purerat testis leydig cell,cultured with bovine serum in DEME/F12 with human recombinant IL-1β,then extractedtotal cell RNA according to the Chomcynski and Sacchi’s,finally analized by Northern blot.It was found thatIL-1β increases in IL-1α mRNA expression dose dependent in Leydig cell.Lipopolisaccharide(LPS)also stim-ulated IL-1α mRNA experssion,but phorbol ester had no effect.The results indicate that Leydig cells are apotential source of IL-1 which has both autocrine and paracrine effects.
