苏云金杆菌10亚种的质粒DNA图谱分析和杀蚊晶体毒素蛋白基因在质粒上的定位 被引量:1

摘要 对苏云金杆菌(Bacillus thuringiensis,简称B.t.)血清型H_1-H_9和H_(14)10个亚种的12个菌株质粒DNA图谱作了琼脂糖凝胶电泳分析。质粒DNA分子量在3.3~150MD之间,其中苏云金亚种(B.t.subsp.thuringiensis)HD-2菌株、戈尔斯德亚种(subsp.kurstaki)HD-1菌株和鲇泽亚种(subsp.aizawai)含有质粒10个;以色列亚种(subsp.israelensis)质粒6个;苏云金亚种berliner菌株和莫里逊亚种(subsp.morrisoni)质粒6个;质粒5个的有戈尔斯德亚种的HD-73菌株和多窝亚种(subsp.tolworthi);猝倒亚种(Subsp.sotto)和有蜡螟亚种(subsp.galleriae)分别有3和4个;而幕虫亚种(subsp.finitimus)和亚毒亚种(subsp.subtoxicus)有质粒2个。以色列和莫里逊亚种编码的杀蚊毒素蛋白基因分别定位于75MD和98MD的质粒上,它们与苏云金杆菌PG-14的cryIVD基因的同源性大于85%。 The 12 strains of Bacillus thuringrensis have been examined for the presence of plasmid DNA by argarose gel electrophoresis, which show that each strain contained a different complex array of plasmid. The plasmid sizes ranged from app-roximatety 3. 3 to 150MD and the number ranged from 2 to 10.Southern hybridization experiments showed highly homology (>85%) amoung the cryIVD gene probe from B. t. morrisoni PG-14 strain and the 75MD plasmid in subsp. israelensis or 98MD plasmid in subsp. morrisoni.
