摘要
采用逐级高渗盐溶液处理小鼠宫颈癌细胞,获得结构似花环状的宫颈癌影细胞即膜抗原。形态学观察显示该影细胞形状、大小均与宫颈癌细胞相似,核为空泡状,其胞膜连续完整,与核膜、胞浆组成一个空泡状的影细胞。该影细胞已失去分裂增殖能力。用其免疫小鼠可诱导显著的细胞和体液免疫反应,提示该影细胞保留了原有的肿瘤免疫原性,用该影细胞预先接种免疫小鼠可有效地抑制宫颈癌细胞的攻击力,该项研究对于宫颈癌疫苗的研制具有重要意义。
xtraction of membrane antigens on the uterine cervical cancer(U14)cells withhypertonic NaCl solutions left structurally intact entities called U14 ghosts.They were notbroken during extraction. Under light and electron microscope,the nucleus of U14 ghostpossessed approximatelly the some size and shape as before extraction but appeared to be al-most empty.There was a nueleolus─like body in some U14 ghosts,and sometimesfilamentous structure was present in the nucleus.The cytoplasm was less dense than that ofU14 cells.The plasm membrane of U14 ghosts was intact and complete,not broken,andformed a wreath─like structure with nucleus membrane。Lymph nodes,spleens thymuses ofthe mice treated with U14 ghosts apparently increased in weight,and histologically appearedapparent cellular and humoral immune responses.Speciflc antibodies to U14 ghosts in thesera of experimental animals were detected by indirect immunoenzyme test.U14 ghostssignificantlly prolonged the survival of tumor─bearing animals.According to these experi-mental resuIts,it can be concluded that U14 ghosts retain membrane antigenicity of U14 cellsand possess their strong immunogenicity。
